El premi Tony a la millor interpretació d'una actriu protagonista en una obra de teatre és un honor lliurat als premis Tony, una cerimònia establerta el 1947 com a premis Antoinette Perry a l'excel·lència en teatre. El premi s'atorga a les actrius per papers principals de qualitat en una obra de Broadway.[1] Tot i que el premi es va lliurar per primera vegada el 1947, no hi va haver candidats anunciats fins al 1956. Hi ha hagut dos empats en aquesta categoria i un de tres.
